I've heard some bad things about mykinos another island in greece. Apparently there is a large gay population there and they are not afraid to try and pick you up or make inappropriate comments to you or your wife. It may seem funny for a bit but I'm sure it gets old really quickly especially if you're there with your wife.
Santorini is great! Make sure you bring your camera because the picture opportunities are amazing there. While I was there I rented a little quad that didn't go very fast but it was tons of fun either way. I'd suggest renting one and going for a tour/ getting lost on the island. They aren't expensive and you get them all day long. As far as I know there are no rules for them because I was all over the road and no one said anything. I didn't even get one local giving me the evil eye. We were practically drinking while driving them down the roads.
I also visited Ios which is another island in greece and it was more of a party island. The bars never really closed and everyone drank every night all night. If you want to get drunk with a bunch of australians in their mid to high twenties this would be the place to go haha. Personally I had the time of my life there but it would definitely be different if you are with your wife. Thats about all the advice I can think of right now. If you have any questions feel free to ask.

Thanks for the heads up on Mykinos. Sounds like the weekends I go with wifey to West Hollywood, or my neighbors house....oy.
Perfect. This is what my wife was leaning towards.
We were also considering going to Italy or somewhere in the med but I don't want to spend whole days traveling if possible.
PM coming.

Well I might as well for my first post...lol. I traveled to Athens last May for a graduation gift and my girlfriend who is Greek had a fun time showing me around. She lived there her whole life and her sister lives in Athens.
Her family has a nice cluster of property and she can tell you a lot about Greece if you wanted. PM if you are you interested.
PS We are going in Sept/Aug for a wedding and will probably be there about 3 weeks.

Hi, another first post here. I grew up in Greece. The answer to your question depends on what you're looking for. However, here's a suggestion for a nice balance.
Stay in Athens for at least 2 nights/ 2 full days (in other words, excluding travel) when you are leaving Greece. It is best when you are leaving because this ensures you're on the mainland a few days before your scheduled flight. Occasionally you can get stuck on an island because the winds are too high to sail and in Greece you should always leave room for the unexpected.
Stay in the city center; I liked Grand Bretagne. Visit the Parthenon, stadium, etc., and Monastiraki/Plaka in the same day. Have dinner there again around 10p and just just walk around. The next day get a taxi out to Vouliagmeni for a swim and then go to Glyfada for lunch. Bonus trip out to Sounion for the sunset but it is far and you'll be back at the hotel around 11p. You can dine at one of the Fish Taverns on the return trip. Or, if you skip Sounion, then dine again in Plaka at a different place.
Santorini is an excellent choice of islands. Crete is another great destination especially the Chania/Xania area. If you really want to see amazing beaches and postcard-worthy island scenery check out Milos, stay at Psaravolada Hotel. If you go, rent a small car and take one of the smaller boat island tours, instead of the big boats. It's not quite the party island but there are a few fun bars and some good restaurants.
Two islands and Athens is more than enough to see. Intra-island hopping takes too much time for two weeks IMO so no more than two is best.
Check out A Greece Travel Guide: Matt Barrett's Guide to the Greek Islands, Athens and Mainland Greece as well for a laid back perspective. No affiliation. I just find it thorough and helpful even for me.
