A6, it has better features, drives better, and is a better looking car IMO.

A6, brother's wife had the E and hopped out real quick...into a Cayenne S.

You really have to drive 'em and get the feel of both, because it has a lot to do with preference between these two.
IMO...
Exterior: Benz
Interior: Audi
Ride: Audi has a sportier ride, benz is more comfort oriented.
I've driven both cars a bit.
My mother has a 2008 E-350 she bought brand new. The car has had no problems whatsoever in the four years she has had it. It just won't do it for me because of a few things: It's underpowered, this car really NEEDS the V8. For such a large car it could feel more roomy inside, it doesn't have a lot of room on the passenger side foot well because of the 4-matic. I hate the transmission, its notably irritating and clicks off shifts when it's not my intention. Finally, the steering is just plain truck-like. Other than that, it rides really smooth. The brakes are good, and the electronics work seamlessly together (Navigation, bluetooth, cruise control, in-dash display etc. etc.). If the transmission doesn't bother you, this car really makes long rides comfortable, and would make a solid daily driver.
A colleague of mine has an A6 3.0T and I've driven it a few times as well, but not as much as I've driven the E-Class. It looks really good on the outside, I just think the benz looks better. The interior is, in typical Audi fashion, class leading. The inside is pretty spacious per volume. My friend was one of the few that had problems stemming from a faulty water pump design, but I hear this issue has since been rectified. The A6 steering feels a little tighter than the benz, but still too little loose for my preference. I'm not crazy about the V6 supercharged engines, but they're quick. The transmission feels better too.
I feel like the A6 has more of a fun, sportier drive, but for some reason the Mercedes E-Class still feels like the more relaxing cruiser.You see my Louis chucks, Louis bag, Louis frames, Louis belt
